High-Performance Liquid Chromatography (HPLC)

HPLC is the standard method for determining peptide purity. The sample is dissolved in a mobile phase and passed through a reversed-phase column under high pressure. Components separate based on their hydrophobicity, each with a distinct retention time. Purity is calculated as the percentage of the main peak area relative to all detected peaks. A high-purity peptide shows a single dominant peak, while additional peaks indicate byproducts or contaminants .

Sample requirement: 1–2 mg, ideally lyophilized. If in solution, please provide dissolved in water.

Liquid Chromatography-Mass Spectrometry (LC-MS)

LC-MS combines the separation power of HPLC with the molecular identification capability of mass spectrometry. This technique confirms the molecular weight of peptides and identifies mass-shifted impurities such as truncations, deletions, or chemical modifications that may occur during synthesis — impurities that are often difficult to detect by HPLC alone .

LC-MS is particularly suited for small to medium-sized peptides and complex peptide mixtures. It offers high sensitivity and tolerance to common contaminants such as salts and buffer components, making it ideal for rapid, accurate mass profiling of peptide samples .

Sample requirement: 0.5 mg minimum.

Deliverable: Mass spectra covering the range including the target mass.

Certificate of Analysis (COA)

Every analysis result is issued as a formal Certificate of Analysis (COA). A COA is a batch-specific document of conformity that lists test results against approved specifications for the product, intermediate, raw material, or component. It includes traceability information, test methods, acceptance criteria, sampling plans, and authorized signatures .

For peptides and pharmaceuticals, the COA typically includes identity confirmation, purity quantification, related substances, residual solvents, moisture content, and physical properties such as appearance and pH .
